Overview

Protein interactions are the physical associations between proteins that mediate most cellular processes, including signal transduction, the assembly of macromolecular complexes, and the allosteric and regulatory control of enzyme activity. The specificity and stability of these interactions are governed by domain architecture and the chemistry of binding interfaces, exemplified by elastic immunoglobulin domains such as the NH2-terminal Ig modules of insect projectin and by the structural consequences of disease-associated variants in proteins like BRCA1. Interactions are mapped experimentally through affinity purification coupled to tandem mass spectrometry, including optimized workflows and quantitative analyses such as ZSPORE in mammalian cells, and are increasingly predicted and characterized computationally through in-silico modeling and molecular docking, as in studies of marine antifungal peptides inhibiting Candida proteins and in computer-aided drug design. Reconstructing these relationships into interactome networks clarifies how signaling axes operate, illustrated by senescence-associated satellite-cell signaling through the FGF2–FGFR1–p38MAPK pathway with its Sprouty1 and microRNA regulators, and connects molecular mechanism to disease, as in coronary-disease variant analysis and the cellular and evolutionary biology of RNA-binding proteins such as RBM45. The journal publishes peer-reviewed research on affinity-purification mass spectrometry, structural and domain analysis, signaling-axis biology, and in-silico interaction prediction and drug discovery.
