Overview

Protein expression and analysis encompasses the production of proteins through transcription and translation together with the experimental study of their abundance, identity, structure, post-translational modifications, and function. Proteins may be expressed in their native context or produced recombinantly in heterologous systems for detailed characterization, and their analysis bridges single-protein studies and proteome-wide surveys spanning whole tissues and organisms. The analytical toolkit centers on gel- and mass-spectrometry-based proteomics, including quantitative strategies such as metabolic labeling with 15N SILAC and label-free ion-current-based approaches that measure relative protein abundance across conditions. These methods are complemented by gene and transcript expression analysis and by bioinformatic interpretation that links sequence variation and regulatory elements to expressed protein function. Applications are broad: profiling the alveolar macrophage proteome in transgenic surfactant-protein models, dissecting butyrate-induced apoptosis resistance in colorectal cancer, characterizing regulators such as optineurin (FIP-2) and their downstream chemokine and kinase signaling, and interrogating disease-associated variants in coronary disease, EPAS1, and microglial loci such as TREM2 and MS4A6A. Comparable approaches extend to plant systems, as in MLO-mediated powdery-mildew responses in Fragaria, and to therapeutic response, as in proteomic analysis of lapatinib sensitivity. The journal publishes peer-reviewed research on mass-spectrometry and quantitative proteomics, expression analysis, and computational interpretation.
