Overview

Pathogenesis is the biological process by which a disease originates and develops, encompassing the sequence of cellular, molecular and physiological events that lead from an initiating cause to the signs, symptoms and tissue changes of the disorder. It addresses aetiology, the mechanisms of injury and dysfunction, and the progression of disease, integrating molecular biology, genetics, immunology and pathology to explain how causative agents, genetic alterations and host responses interact. Elucidating pathogenesis is central to identifying diagnostic markers, therapeutic targets and strategies for prevention. Research relevant to this area includes bioinformatic analysis of disease-associated SNPs and genes implicated in the pathogenesis of atherosclerosis; haematological disturbances and their pathogenesis in dengue haemorrhagic fever; the histologic, genetic and molecular characteristics of meningioma pathogenesis and progression; the molecular and metabolic pathogenesis of familial combined hyperlipidaemia and metabolic syndrome; the role of amino acids and their derivatives in liver-disease pathogenesis and treatment; an introductory review of biological networks underlying disease; the biology of the novel coronavirus; the interaction of frailty with the immune system; and label-free proteomic analysis of a parasite. Studies of contrast-agent toxicity and trace-element content in thyroid disease further illustrate mechanism-focused work. Across these contributions the field reconstructs the molecular and cellular pathways that drive disease, linking causal factors to clinical manifestation and to opportunities for intervention.
