Overview

COVID-19 diagnosis is the process of confirming SARS-CoV-2 infection through clinical assessment and laboratory or imaging investigation. The reference standard is detection of viral RNA by reverse transcription polymerase chain reaction (RT-PCR) on respiratory specimens such as nasopharyngeal or throat swabs, which identifies active infection with high sensitivity; rapid antigen tests offer faster point-of-care detection at lower sensitivity, and serological assays detect prior exposure through antibodies. Diagnostic workup integrates clinical evaluation of symptoms and exposure with these tests, and chest computed tomography contributes by characterising the pattern and extent of pulmonary involvement in suspected or confirmed cases. Laboratory parameters, including white-cell and lymphocyte counts and inflammatory markers such as ferritin, support assessment of disease activity and prognosis rather than primary diagnosis. Comparative evaluation of commercial RT-PCR assays and prognostic scoring tools refines diagnostic accuracy and risk stratification, while mathematical modelling supports epidemic prediction and control.
