Overview

Enzyme purification is the process of isolating a single enzyme from a complex biological mixture so that it can be studied or used in a pure, active form. Because cells contain thousands of different proteins, purification relies on a sequence of separation steps that exploit differences in size, charge, solubility, hydrophobicity, and binding specificity, typically combining techniques such as precipitation, centrifugation, and various forms of chromatography while monitoring enzyme activity at each stage. The goal is to obtain a preparation with high specific activity and minimal contaminating proteins, enabling accurate characterisation of the enzyme's structure, kinetics, and function, as well as its application in research, diagnostics, and industry. Research published in the journal Enzymes illustrates these aims, including the identification and characterisation of a novel exo-acting glucanase and xylanase from Trichoderma reesei and the optimisation, production, and purification of laccase from a Bacillus species. Other work characterises hydrolytic and oxidative Enzymes and their catalytic properties, reflecting the journal's focus on enzyme discovery, production, and mechanism. This page gathers peer-reviewed, open-access research relevant to the isolation, characterisation, and application of Enzymes.
