Overview

Acid phosphatase is an enzyme that catalyzes the hydrolysis of phosphate monoesters, liberating inorganic phosphate from organic substrates, and is defined by its optimal activity under acidic pH conditions, which distinguishes it from the alkaline phosphatases. It occurs widely across bacteria, plants, and animal tissues, existing in multiple isoforms with tissue-specific distribution; in humans, prostatic and tartrate-resistant forms are well characterized, the latter being a recognized marker of osteoclast activity and bone resorption. Functionally, acid phosphatases contribute to phosphate metabolism and nutrient mobilization, to lysosomal degradation, and to physiological processes spanning digestion and mineral handling. Their activity serves as a biochemical indicator in clinical and experimental settings: changes in enzyme levels accompany disturbances of bone metabolism, including experimental osteoporosis, and shifts in tissue acid phosphatase reflect organ responses to toxic or pharmacological exposure in blood, liver, and muscle. As a measurable enzymatic parameter, acid phosphatase is incorporated into biochemical panels and proteomic analyses that assess tissue status and treatment effects. Because its expression and activity vary with cellular turnover and pathological state, the enzyme functions both as a participant in normal metabolism and as a sensitive biomarker. Research addresses its enzymatic properties, its role in bone and tissue metabolism, and its use as an indicator of physiological and toxicological change.
