Overview
Protein sequencing is the process of determining the order of amino acid residues that make up a protein or peptide. Because the amino acid sequence dictates how a protein folds into its three-dimensional structure and therefore how it functions, sequencing is a foundational technique in biochemistry, molecular biology, and proteomics. Classical approaches such as Edman degradation read residues from the end of a peptide chain, while modern high-throughput work relies heavily on mass spectrometry, which identifies and sequences peptides by their mass-to-charge ratios. Protein sequences can also be inferred indirectly from the DNA sequence of the genes that encode them. Sequencing is used to identify and characterize newly discovered proteins, to detect post-translational modifications, to confirm the identity and purity of recombinant proteins, and to study how variations in sequence relate to disease.
