Overview
Protein-protein interactions (PPIs) are the physical contacts established between two or more protein molecules, and they underlie virtually all cellular processes, from signal transduction and metabolism to gene regulation and structural organization. Mapping and understanding PPIs is central to molecular biology because most proteins function as part of complexes and networks rather than in isolation, and disruptions to these interactions are implicated in many diseases. The topic includes both experimental approaches—such as affinity purification coupled with mass spectrometry—and computational and bioinformatic methods used to predict, model, and analyze interactions and their networks. Related areas include proteomics, structural biology, interaction databases, and the use of interaction data in drug discovery and the interpretation of disease-associated genetic variants. The journal publishes research touching on these dimensions, including optimization of affinity-purification mass spectrometry in mammalian cells, bioinformatic analysis of disease-associated genes and proteins involved in atherosclerosis, the role of bioinformatics in biotechnology and disease diagnosis, the Human Proteome Project and its bioinformatics status, structural and functional analysis of protein variants of uncertain significance, in silico inhibition of microbial proteins by antimicrobial peptides, and computer-aided drug design. This body of work illustrates how protein-protein interactions are studied through combined experimental, structural, and computational strategies relevant to biotechnology and biomedicine.
