Overview

Chromatography is a family of analytical and preparative techniques used to separate the components of a mixture based on their differential partitioning between a stationary phase and a mobile phase. As analytes move through or across the stationary phase, differences in properties such as polarity, size, charge, or affinity cause them to migrate at different rates, achieving separation that enables identification and quantification. Major formats include thin-layer chromatography, widely applied to phytochemical profiling and the separation of pharmaceutical compounds, and high-performance liquid chromatography, frequently coupled with mass spectrometry to provide sensitive and specific detection. Liquid chromatography-tandem mass spectrometry is a mainstay for determining drug and metabolite concentrations in biological fluids, supporting bioequivalence and pharmacokinetic studies, and for analyzing complex matrices in food safety and natural-product chemistry. The technique is essential across biochemistry, pharmaceutical analysis, food science, and synthetic chemistry, where it is used to isolate and characterize secondary metabolites, verify the composition of formulations and supplements, and detect adulterants.
