Overview

Non-steroidal anti-inflammatory drugs (NSAIDs) are a class of pharmaceuticals that relieve pain, reduce fever, and suppress inflammation by inhibiting cyclooxygenase (COX) enzymes, thereby blocking the synthesis of prostaglandins from arachidonic acid. Two principal isoforms are targeted: COX-1, which supports gastric mucosal protection and platelet function, and COX-2, which is induced at sites of inflammation. The balance of inhibition between these isoforms shapes both efficacy and the characteristic adverse-effect profile, including gastrointestinal, renal, and cardiovascular risks, and underlies the development of preferential and selective COX-2 inhibitors intended to spare platelet aggregation and gastric integrity. Research in this area addresses the comparative pharmacology of selective and non-selective agents, hypersensitivity and cross-reactivity among COX-2 inhibitors in susceptible patients, the analgesic and anti-inflammatory evaluation of novel chemical derivatives and plant-derived agents, and the potential role of these drugs in settings such as colorectal cancer chemoprevention and the management of arthritic and post-surgical pain. Bioanalytical methods supporting pharmacokinetic and bioequivalence assessment of related compounds also feature. NSAIDs matter because of their extensive clinical and over-the-counter use and the need to optimise the benefit-risk balance. The journal publishes peer-reviewed research on the pharmacology, analysis, and clinical application of these agents.
